Melatonin: Chemically known as N-acetyl 5 methoxytryptamine, it is a natural compound produced by a gland located at the base of the brain and known as the epiphysis. Its production in the human organism follows a very precise rhythm, called the circadian rhythm, which has a maximum peak between 00.00 and eight in the morning, and which seems strongly inhibited by sunlight.
Although taken through various foods (cocoa, rice and other vegetables), dietary melatonin is not able to significantly increase the plasma levels of the hormone, and for this reason a specific supplement is used. Taken in the form of capsules, in fact, melatonin is totally absorbed in the intestine for doses up to 2.8 mg, but largely metabolized in the liver, where it transforms into its inactive metabolite 6 sulfotoxymelatonin, so much so that its bioavailability decreases up to at 15%. The maximum plasma concentration is reached on average after 3 hours, even if this time interval seems to be seriously influenced by the nutritional status of the subject. The half-life of melatonin does not normally exceed 4 hours. Through the bloodstream, bound to plasma proteins for about 60%, melatonin is able to diffuse freely and bind its receptors (MT1, MT2 and MT3) present mainly in level of the central nervous system.
Biological role: several experimental evidences and numerous studies in this regard, show that melatonin has a:
- Regulatory effect on the sleep-wake rhythm: through the hypnotic, sedating and inducing effect of sleep;
- Antioxidant effect: able to prevent damage induced by both reactive oxygen and nitrogen species. In this case, however, the amount of melatonin involved is irretrievably lost, since there are no effective reduction reactions. However, its antioxidant action seems quite powerful, since it is also guaranteed by the subsequent metabolites.
- Protective and anticarcinogenic effect: melatonin appears to be involved in the protection of mitochondrial DNA, protecting it from the mutagenic action of various compounds.
- Immunostimulating action: although not fully characterized, first evidences show a significant enhancement of the immune response.
Melatonin and sport
Although not particularly widespread among athletes, in recent years melatonin has also found various applications in sports, where it can be used for:
- Guarantee an antioxidant effect, and protect the organism from oxidative damage;
- Strengthen the immune defenses in athletes subjected to strenuous training and at risk of overtraining syndrome;
- Improve the hormonal profile.
In this regard, worthy of note are some studies in which it is shown:
- the metabolic effect of melatonin: able to reduce the production of lactate during physical activity;
- the hormonal effect: where there is an increase in GH levels which occurs at doses of 0.5mg and 5mg of melatonin per day.
Recent evidence, based on the observation of a hypothermic effect, suggests a potential usefulness of melatonin in improving performance in the presence of high temperatures.

Melatonin is currently used successfully in all various sleep disorders, such as jet lag syndrome. Numerous studies, however, are underway to test its effectiveness also for some neurodegenerative diseases, such as Alzheimer's and amyotrophic lateral sclerosis, pathologies induced by oxidative damage, cardiovascular diseases and more.
In sports, some studies demonstrate the efficacy of melatonin in improving hormonal secretion and assisting the body's antioxidant function. Also in this case the dose recommended by the manufacturer seems to be the most valid, even if more than one study shows how an improvement in the nocturnal secretion of GH can be obtained even at significantly lower doses, equal to 0.5 mg.
Given the pharmacokinetic properties of the supplement, it would be advisable to take it before going to bed, so that its maximum peak corresponds to the normal endogenous secretion.
