Melatonin: also known as N acetyl 5 methoxytryptamine, it is produced at the level of the epiphysis starting from the amino acid tryptophan. Its synthesis is not only regulated by the availability of the substrate, but is affected by the activation of a key enzyme known as serotonin N acetyl transferase. The control of this enzyme is mediated by a series of biological phenomena, not yet fully known, which see the association between external stimulus and biological function. In fact, the presence in the pineal gland of cells similar to retinal photoceptors makes it particularly sensitive to light, so much so that it can modulate its activity based on the light / dark cycle. The electromagnetic wave (light), hitting these photoceptors, induces functional changes in the cell, which lead to an increase in cyclic AMP with consequent inactivation of the enzyme Serotonin N-Acetyl Transferase, and a reduction in melatonin synthesis. It can therefore be easily deduced that the production and secretion of melatonin is maximum at night and is zeroed during the day. The maintenance of this cicardian rhythm guarantees the correct functioning of the organism and in particular of the clock that regulates sleep and wakefulness.
The sedative and sleep-inducing effect is guaranteed by the presence of receptors for melatonin, present at the hypothalamic level and capable, once activated, of inducing the hypnotic and sedating effect that characterizes this molecule.
However, recent evidence underlines how melatonin is particularly active also in other parts of the body, mediating different biological actions, such as:
- Antioxidant effect: it has been shown that melatonin in primis, but also the subsequent 4 metabolites that derive from its oxidation, can protect the organism from the oxidative insult induced by reactive oxygen and nitrogen species. In this case, however, the absence of enzymes capable of reducing these species results in the net loss of the amount of melatonin involved in the red-ox process.
- Immunostimulating effect: melatonin has been shown to significantly improve the antibody response and the cytotoxic response, also enhancing the secretion of pro-inflammatory cytokines. This action could be dangerous in case of patients suffering from autoimmune or allergic diseases.
- Protective effect: several studies support the protective action against mitochondrial and nuclear DNA by mutagenic and potentially carcinogenic agents. Initial evidence also suggests an anti-neoplastic action, observed however exclusively in vitro and on animal models.
- Metabolic effect: also in this case, some studies conducted in vitro and on animal models, have suggested an important metabolic action, which seems to be associated with the MT1 receptor and is expressed through an improvement in insulin sensitivity, especially in obese, diabetic and fed models. diets high in fat.
In sports, melatonin has few fields of application, given the presence of other more effective and targeted supplements; however, it is used by some athletes in an attempt to improve hormone secretion, protect the body from overtraining syndrome, and strengthen the pull of antioxidant defenses.
The studies, however, do not seem very much in agreement and in the sports field there is no such experimentation as to allow conclusions to be drawn; despite this there are some works in which, following the administration of melatonin, there is an improvement in the nocturnal secretion of GH, an improvement in the antioxidant capacity and a protection of the immune system. However, it should be reiterated that these advantages would be expressed through the reproduction of the normal circadian rhythm, avoiding subjecting the body to further stress and hormonal deregulations.

Take 4 drops of liquid equal to 1 mg of melatonin, using the dropper contained in the package, with water or juice, in the evening 20 minutes before going to bed.

The various studies, which for several years have recommended minimum dosages of 3 mg per day, have been joined by new studies that show the same results even for significantly lower dosages, even equal to 0.3 mg. The advantage of the liquid form compared to other supplements in capsules or tablets, does not materialize with the improvement of the kinetic properties, which remain unchanged, but in the simpler intake and above all in a better modulation of the dosage, considering that on average per drop there would be about 0.25 mg of product.
Also in this case, the supplement should be taken before going to bed, in order to preserve the normal rhythm of endogenous secretion, reducing the "working" weight of the epiphysis.

Several studies seem to agree on the greater effectiveness of the synergy when taken together with zinc. A generalized improvement is in fact obtained both on the strengthening of the immune defenses, and on the antioxidant effect and on the metabolic aspect, improving the sensitivity to insulin.
Some studies show that in case of prolonged intake of zinc and melatonin, supplementation with magnesium is also necessary.
The validity of the simultaneous administration of melatonin and vitamin B6 is still under observation, even if the first works show an improvement in the endogenous synthesis of this hormone, with an enhancement of the biological effects related above all to the modulation of the immune and hormonal activity.

Although recent studies have confirmed the high tolerability and safety of the active ingredient, there are cases in the literature in which, even at a dose of 3 mg / day, nausea, irritability, nightmares and vascular alterations have been observed.
Furthermore, by acting as a mild hypnotic and sedative, melatonin may interfere with the ability to concentrate. By enhancing the immune response, the substance could also worsen the conditions of patients with allergic or autoimmune diseases.

The product is contraindicated in cases of renal or hepatic pathology, cardiovascular disease and / or hypertension, allergies and autoimmune diseases, during pregnancy, during lactation, under the age of 12 and in adolescents not yet trained.
